The principle of embedding is to immerse certain special supporting substances into the interior of the tissue block, utilizing the physical properties of the supporting substances, such as transforming from liquid to solid, to ensure that the entire organization has a uniform and consistent solid structure and sufficient hardness, in order to facilitate the use of slicing mechanisms to obtain extremely thin slices. The most commonly used embedding method in histological slicing technology is to use paraffin as the embedding agent, and place the tissue block soaked in paraffin to make a wax block.
Due to the softness or uneven hardness of the tissue, it is difficult to make slices with uniform thickness using conventional techniques when making slices or ultra-thin slices. Therefore, it is necessary to immerse certain substances inside the tissue, and the entire tissue will harden equally. First, melt paraffin is injected into the embedding bracket (mold), and then use tweezers to remove the tissue block that has been soaked in wax from the dehydration box, place it in the center of the embedding bracket, and place it on a small cold table, Gently press the tissue block with tweezers to achieve flatness. Cover the bottom of the dehydration box, add paraffin, and move to the freezing table. After freezing, remove the tissue wax block for cutting.
