Dyeing principle

The Pap staining solution contains dyes such as hematoxylin, orange yellow, eosin, bright green, and Bismarck brown. Among them, hematoxylin is an alkaline dye that can bind to the main chemical component deoxyribonucleic acid in the cell nucleus, resulting in a purple blue color.
